Jet lag is what happens to your body after crossing multiple time zones quickly i.e. in a jet. Basically, your body is used to eating, sleeping (and going to the toilet!) at certain times of the day. Your body will take some time to adjust to the shift in time, and getting used to sleeping, eating, (and pooing!) a few hours later or earlier than normal. I don't think it's that bad, just annoying sometimes. I recently flew from London to NYC. The first 24 hours were weird; I woke up at 4am because my body was convinced it was 9am (London time). I was in the Met Museum of Art at 4pm but was almost falling asleep because my body thought it was 9pm (London time). Quite cool really and a strange sensation, and it soon wears off by day 2 or 3. Try and adopt the local time as soon as possible.

Jetlag happens because of the time zone change.
It affects everyone differently..When you come to Europe, your biological clock will be working as if you were still in NC. There is nothing special you can do about it, try to adapt yourself to the time zone in Prague.
Don't be scared, some people don't even feel it..
